Sequencing of Barley ESTs
The shortest path to gene discovery, mapping of genes of choice, and analysis of plant traits, which are likely to be multigenic, is the large-scale sequencing of EST libraries. The ESTs (Expressed Sequence Tags) are single-pass sequences from cDNA libraries. Mechanization of the colony picking, PCR, and sequencing steps makes the process very rapid and efficient. In addition to the Finnish importance of barley, barley EST data is relevant to the closely related wheat and rye without the hexaploid complexity of wheat. We are currently running a barley EST project funded by the Finnish technology development center TEKES with the goal to produce sequences of at least 50000 expressed genes from resting, developing, and germinating grains, leaves, roots, and various infected tissues. |
